Generation of neuronal models from patient-derived induced pluripotent cells is often hampered by the lack of maturation and connectivity of the differentiating neuronal cell types, reducing the chances for a relevant in vitro phenotype and read-out. The lecture will discuss how three dimensional neuronal cultures are suitable to increase the complexity and promote maturation of the cells, and to what extent they might provide a remedy to some of the problems above.
